V tem delu smo opisali pripravo modela GBM sferoidov in postavitev modela za študij invazivne subpopulacije tumorskih celic. Uspeli smo separirati to invazivno celično subpopulcijo iz organotipskih primarnih kulturi GBMov iz bolnikov in določiti delni protezni profil, ki je pokazal na ključno vlogo katepsina B ki se v tem invazivnem celičnem fenotipu verjetno aktivira postranslacijsko. V nadaljevanju dela bomo ta model lahko uporabili tudi za proučevanje matičnih celic tumorja v invaziji in apoptozi ter možnosti inhibicije s sintetskimi inhibitorji - kar je predmet tega projekta.

In this work, two proteolysis related markers were investigated in two cohorts of glioma patients: plasminogen activator and lysosomal cathepsin B. Although plasminogen activator- dependent proteolytic system plays a role in the progression of various tumour types, it seems that in glioblastoma cathepsins B is more relevant with statistically stronger prognostic impact. This is most likely due to its involvement in GBM invasion and migration, as we have confirmed during the first year of the project in vitro in glioblastoma primary spheroid culture and in the model cell lines.
