The resuts of ou collaboration with industry is the new method of detection of phytoplancton organisms using fluorescence with the use of directed flow in the chamner. This was submitted in 2016 to Europen Patnet Office (EPI) in 2016, under the differenet title as te underlaying Slovenian patent, and the ttile "Method and system for simultaneous detection of nanparticels concentration in suspension and ther morphological and physiological characterstics (EP 3 073 246 A1).

COBISS.SI-ID: 3783759Proteases, including lysosomal cathepsins, are involved in many processes in cancer progression. Only recently, cathepsin K (CatK), originally reported as a collagenolytic protease produced by osteoclasts, appeared to be overexpressed as well in various types of cancers, including brain tumour glioma. In this review, the physiological functions of CatK are presented and compared to its potential role in pathobiolology of processes associated with tumour growth, invasion and metastasis of cancer cells and their interactions with the tumour microenvironment. CatK activity is either indirectly affecting signaling pathways, or directly degrading extracellular matrix (ECM) proteins. In glioma CatK possibly regulates cancer stem-like cell mobilisation out of their niches and modulates recently found physiological CatK substrates, including chemokines and growth factors. Finally, the application of CatK inhibitors, which are already in clinical trials for treatment of osteoporosis, has a potential to attenuate cancer aggressiveness.

COBISS.SI-ID: 283891456Mesenchymal stem cells (MSCs) are widely use in cell therapy and regenerative medicine. To obtain the large numbers of cells that are necessary for clinical applications, MSCs need to be isolated from the tissue source and expanded in vitro. Adipose tissue (AT) is the most convenient origin for MSCs, and thus we explored the anatomical location, such as hips and abdomen as the harvests sites for AT-MSCs), and examined the key parameters for AT-MSC expansion on micro carries in spinner flasks. Besides several growth parameters we also proved better adhesion Cytodex-3 and Cytodex-1 and expansion of AT-MSCs in spinner flasks with rotation than without it. The differentiation of AT-MSCs from hips are more prone to differentiation into bone and cartilage cells, whereas highest quality of the cells was achieved from abdominal fat. The study on paracrine effects of MSC on glioblastoma stem cell (GSC) lines showed that MSC conditioned media factors impaired proliferation of GSCs via Cyclin-D1-mediated cell-cycle arrest, resulting in induction of senescence. This appeared to be mediated via signaling through two tumour-suppressor pathways: p53-p21-pRB and p16- pRB. MSC paracrine signals also increased the sensitivity of GSCs towards the chemotherapeutic temozolomide, and induced astrocyte-directed differentiation of GSLCs by down-regulation of Sox-2 and Notch-1, and induction of vimentin and GFAP gene expression. The impact of this work is to show the anti-tumour activity of MSC, targeting cancer stem cells and on the ways to harvest more MSC preferably from abdominal fat tissues

COBISS.SI-ID: 852855Tamara LAH TURNŠEK Lecture at University of Louisville, Department of Microbiology & Immunology, Immunology Discussion Club Meeting Series, Louisville, KY, October 18, 2016. In this lecture the heterogeneity of U87, U251, and U373 glioblastoma (GBM) cell lines is decribed as, this being important when these are used in vitro cell models to determine the inter-cellular communication. Here we used transctrptomic approach to determine the GBM cells molecular fingerprints, showing that U87-cells have lower the genomic stability of U373 (U251) cells, without affecting their proliferation, while oppositely, U87 cells showed increased genomic stability, decreased proliferation rates and increased invasion. Indirect and direct mututal co-culturing of U87 and U373 cells showed mutually opposite effects on temozolomide resistance. By defining transcriptional alterations between various GBM cells we providedfor better understanding of the mechanisms of glioblastomaheterogeneity, which shall provide the basis for more informed gliomatreatment in the future.
