With systematic analysis of B-class and C-class ODNs we determined the minimal sequence requirement that efficiently activates human TLR9. The effect of nucleotides and the distance between CpG motives as well as dimer formation on TLR9 activation were analysed. The minimal ODNs with general sequence TCG[T]6-10CG[T]9-19 and length of 21 nucleotides were prepared. Selected minimal ODNs activated synthesis of TNFa, IL6 and IL12 on human cells.

Species-specific minimal sequence motif for oligodeoxyribonucleotides activating mouse TLR9. We determined that for activation of mTLR9 only one CpG motif 4 to 6 nt distant from 5' end of ODN is required. We prepared minimal ODNs with general sequence [T]4–6CG[T]12–21 longer than 20 nt. Selected minimal ODNs induce synthesis of mIL6 and mIL12p40 in mouse dendritic cells, activate B-cells and RAW macrophages. We showed that mouse-specific minimal ODNs fail to induce activation of hTLR9.
