Although cysteine cathepsins were traditionally considered to be involved mainly in bulk protein degradation that takes place within the lysosomes, it has become clear that their proteolytical activity can also influence specific pathological processes such as cancer, arthritis and atherosclerosis. Here, we presented an overview of cysteine cathepsin roles in disease progression and we discussed their potential relevance as prognostic and diagnostic biomarkers.

Cysteine cathepsins are among the major proteases involved in ECM remodeling and their role is not limited to degradation only. In this review we focused on their extracellular functions linked to ECM degradation and discussed their importance in pathologies and treatment of ECM-linked diseasses.
