This review article mainly covers research in the field of cysteine cathepsins, also linked to pathological states. In this regard, a detailed study of the potential role of cysteine cathepsins in cancer is discussed. This article was dedicated to the discovery of lysosomes and Nobel prize winner Christian de Duve.

This review article summarizes the data accumulated in the research of the role of cysteine proteases in genetically engineered mouse cancer models. Moreover, provides some insight on how to address the next preclinical challenges for developing cathepsin inhibition as cancer therapy. It proposes the improvement of inhibitor selectivity and targeted delivery to the tumor microenvironment and investigation of the biological context of the individual factors within the complex proteolytic network.
