In this paper we presented the problem of cytomegalovirus (CMV) promoter, which is one of the most widely used promoters for transgene expression in mammalian cells. The aim of thestudy was to assess the role of methylation, irradiation and treatments with chemotherapeutic cisplatin on the activity of this promoter by using non-invasive in vivo fluorescence imaging. We proved that with radiation and cisplatin activity of CMV promoter is induced. As a control promoter, mammalian inducible promoter of p21 gene was used. From the results, we concluded that the observed variations in CMV promoter activity limit the usefulness of this promoter for gene therapy.
