We demonstrated experimentally that endocytotic vesicles in B16 F1 mouse melanoma cells are electropermeabilized by nanosecond, high-voltage electric pulses. The method could be used to introduce various substances into living cells. In contrast to some previous publications in the field of nanosecond electropermeabilization, we have shown that the permeabilization of internal cell membranes at given electrical parameters is accompanied by a permeabilization of plasma membrane.

We modeled and simulated in SPICE the diode opening switch generators for intracellular membrane permeabilization. To compare the models with real circuits, we have chosen two diode opening switch generators from the literature, one that utilizes linear air-core inductors and another that utilizes saturable magnetic core in their pulse forming network. The results obtained by SPICE simulation are in good agreement with the previously published measurements of real circuits. Our models of electrical elements are thus sufficient to study, analyse or design new diode opening switch generators.
