In this study, electrogene therapy (EGT) as a new therapeutic approach to canine MCTs, was established. Eight dogs with a total of eleven cutaneous MCTs were treated with intratumoral EGT using DNA plasmid encoding human interleukin-12 (IL-12). Intratumoral EGT with IL-12 elicits significant reduction of treated tumors’ size, ranging from 13% to 83% (median 50%) of the initial tumor volume. These data suggest that intratumoral EGT with plasmid encoding IL-12 may be useful in the treatment of canine MCTs, exerting a local antitumor effect.

The review article is a comprehensive review of preclinical and clinical studies on IL-12 electrogene therapy of cancer. The preclinical studies have demonstrated that IL-12 gene elctrotransfer is an effective approach as local or systemic treatment. Translation of preclinical studies into clinical trials in human and veterinary oncology has started with encouraging results that would hopefully lead to further investigation of this therapy, also in combination with other cancer treatment modalities.
