We have developed approach for identification of functional SNPs in cis-elements associated with allele specific expression in disease candidate regions. In this approach we initially correlated our gene expression data we have obtained from lymphoblastoid cell lines from CEPH families to public released International HapMap project genotype data and confirmed genotype-gene expression correlation in blood lymphocytes and tissue biopsies isolated from patients with Inflammatory bowel diseases. Using this approach we confirmed PTGER4 gene as best candidate gene in 5p13.1 IBD candidate region.

COBISS.SI-ID: 13278998We have used IBDQ test to monitor therapy response to new biological drug adalimumab- during 1 year period treatmen of 75 Croh disease (CD) patients. We have collected biochemical and immunological parameters during treatment and genotyped SNPs in selected genes. After 52 weeks treatment 21 (28%) of patients were classified as good, 29 (39%) as moderate responders and 25 (33%) as non responders according.. Identified genotype, gene expression, biochemical and immunological correlations with IBDQ enable us to design optimized pharmacogenomic profiles for personalized treatment
