Nucleic acids as double-stranded DNA, double-stranded RNA and single-stranded RNA are ligands for endosomal Toll like receptors TLR9, TLR3 and TLR7/8. Nucleic acid ligand binds to TLR receptor and triggers activation of innate immunity. Work describes signalisation events of innate immunity and role of nucleic acids and its analogues in the named process. The use of nucleic acids and their analogues in activation of immune response has been discussed.

Inhibition of endosomal TLR receptor activation with antimalarial drugs has been known for several years; however the mechanism of action was not elucidated. In named work the binding of antimalarial drugs on ligands of endosomal TLRs has been demonstrated. We proposed that structure of ligands, nucleic acids changes conformation in the presence of antimalarial drugs, which consequently prevents binding of ligand to the corresponding receptor and its activation.
