In this work we described the preparation of GBM spheroid model and how this could serve for studying the invasive subpopulations of tumour cells. We succeeded in separation of invasive subpopulation from organotypic primary cultures of GBMs (from patients) and determined its proteolytic profile that pointed out on the key role of cathepsin B, which is postranslationally activated in the invasive cells. In the continuation of the work this model will be used to establish tumour stem cells invasion and apoptosis and the effect of synthetic inhibitors- what is the subject of this project.

V tem delu smo raziskali dvoje proteoliznih sistemov v dveh kohortih – primerjalnih skupinah bolnikov z gliomi: plazminogenski aktivator in lizosomalni katepsin B. Čeprav igra plazminogenski aktivator pomembno vlogo pri napredovanju raznih vrst tumorjev, se zdi, da je v glioblastomih bolj pomemben katepsin B, saj ima najmočnejšo statistično napovedno vrednost. To je verjetno pripisati njegovi vlogi v invaziji in migraciji, kar smo potrdili v in vitro poskusih na glioblastomskih primarnih sferoidnih kulturah v času prvega leta raziskav na vsebini tega projekta.
