We developed an assay system with metabolically competent HepG2 cells stably transfected with plasmid containing the fluorescent protein DsRed reporter system under the control of p21 promotor, which has not been used before in such test systems. The obvious advantages of the developed assay are that it is highly sensitive for detection of direct and indirect acting genotoxic compounds, the genotoxic response can be quantified by measuring fluorescence of reporter gene by means of fluorescent microscopy coupled to image analysis system, by flow cytometry or by spectrofluorimeter, and the results are obtained within 48 hours of exposure. As the assay can be performed on the 96-well microplate format only minute amounts of the analyte are needed, it is simple to perform and it has the potential to be developed into robotized high throughput genotoxicity screening assay. In 2011 we got Slovenian patent and an internationaj patent application has been filled in.
