In the clinical practice, 40–60% of rheumatoid arthritis (RA) patients experience adverse events or inefficacy of methotrexate (MTX) treatment. The anti-inflammatory action of MTX is due to its antagonistic effect on folate pathway as well as its effect on intracellular adenosine levels. Polymorphisms in MTX and adenosine metabolic pathways influence the disease and occurrence of adverse events. In clinical practice MTHFD1 G1958A and AMPD1 C34T polymorphisms could serve as markers of MTX efficacy, while ATIC C347G as a marker for occurrence of adverse events.

The exact closed-form solution to the Michaelis-Menten equation is expressed in terms of the Lambert W(x) function. However, the W(x) function is not widely available in curve-fitting software. Based on various approximations to the W(x) function, different explicit equations expressed in terms of the elementary functions are proposed here to fit time depletion of substrate concentration in progress curves using commonly available nonlinear regression computer programs. The results are compared with those obtained by fitting other algebraic equations.

The hydrolysis of butyrylcholine by vertebrate acetylcholinesterase shows time and substrate-concentration dependent decrease in activity. As the substrate depletion results in concave progress curves at low substrate concentrations, this cannot be the reason at higher substrate concentrations. A good explanation was the time-dependent appearance of a non-productive enzyme-substrate complex. The slow establishment of this complex appears to be a rare occurrence of incorrect substrate orientation at the bottom of the active site, blocked by a second substrate molecule bound at the entrance.

We prepared recombinant archiascomycete S. pombe (SpoC12R) with heterologically expressed steroid hydroxylation system (cytochrome P450 and NAD(P)H:cytochrome-P450-reductase) from fungus Rhizopus oryzae. Host organism hydroxylated progesterone more efficiently than previously prepared reccominant fungus S. pombe (SpoC12) only with heterologous cytochome P450 glive R. oryzae. The reason for better activity might be either an improved interaction between heterologous partners or more adequate ratio between the enzymes.

The HIV accessory protein Nef is one of the earliest and most abundantly expressed viral proteins. It is also found in the serum of infected individuals. The mode of exit of Nef from infected cells is unknown. We have shown here that Nef significantly stimulates its own export through the release of exosomes, from all cells examined. Nef was also released with exosomes in the context of HIV-1 infection of primary T lymphocytes. These numerous Nef-containing exosomes, cause activation-induced cell death of target resting lymphocytes in vitro.
