A method for labeling and monitoring of single vesicles in astrocytes was presented. The method was optimized on rat astrocytes. It enables specific labeling of vesicles, which fuse with the plasma membrane. During fusion vesicle components are exposed to the extracellular milieu; this enables binding of specific antibodies to label vesicles.

We presented preliminary data obtained by the new method for labeling and monitoring single vesicle transport in live astrocytes. Vesicles which fused with the plasma membrane and were retrieved back into the cytoplasm were labeled by antiboides against specific proteins in the vesicle lumen of the vesicle membrane. Transport of antibodies labeled vesicles which fused with the plasma membrane is for on order of magintude slower from the same types of vesicles which are trafficking form GA to the plasma membrane.
