In this paper we present development of activity based probes for proteases and we discuss current trends of their application in proteomic analysis of proteases.

Here we present the data showing the influence of cathepsin B on mammary tumor progression and metastasis formation. We have prepared Ctsb-deficient mouse mammary cancer model Tg(MMTV-PyMT). Absence of Ctsb significantly impaired development of high-grade invasive ductal carcinomas and reduced the metastatic burden in the lungs. Cancer cells lacking cathepsin B also exhibited significantly higher resistance to apoptosis induction. Our results indicate an in vivo role for cathepsin B in promoting cellular anaplasia in mammary cancers and proliferation of cancer cells.
