In our study we compared the ability of HPAI isolate H5N1 of avian origin to adopt and grow on different cells cultures of avian and mammalian origin and find out potential molecular changes on main antigenic determinates such as hemaglutinin gene. The growth of the virus was monitored by determination of genome copies by real time RT-PCR and estimation of cythopatic effect (CPE). The results were confirmed that passages on embryonated eggs lead to mutation in hemagglutinine gene meanwhile no mutations were observed after passaging on cell cultures.

So far, only a few data about the resistance of influenza A virus in the environment exist. The purpose of our work was to develop an efficient molecular method for AI virus detection which enables us to trace the virus in complex environmental samples. Different RNA isolation methods were tested and the efficiency of them was determined by an implemented real-time RT-PCR method.
