For large scale Ad vector purification, chromatography using methacrylate monoliths has been found to be effective. Monoliths are characterized by convective mass transport, large channel diameter, high surface accessibility and high dynamic binding capacities for viruses. The aim of this study was exploring the characteristics of adenoviral vector purification using different ion exchange methacrylate monoliths.

Different chemistries of supports and differently prepared crude adenovirus lysates were used in the experiments. It was shown that purity and recoveries were highest using strong anion exchange support. A high dynamic binding capacitiy of viral particles (reaching up to 2 x 10^12 virus particles per ml of support) combined with high flow rates at low pressure drops were further significant characteristics for these monolithic supports, providing efficient down-stream processing in adenoviral vector production.
