HPV L2 colocalises with different subset of exogenously expressed PML isoforms, depending on HPV type. We compared a low risk and high-risk types HPV viruses. HPV-11 L2 colocalises with PML I and II. In addition to PML I and II, HPV-16 colocalise also with PML IV, which is implicated in cellular senescence. HPV-18 L2 colocalise with majority of PML isoforms in large nuclear bodies. Our results confirm that ND 10 structures have different PML compositions and suggest that the viral L2 protein targets only those domains that are important for specific viral activities.

Interestingly, L2 showed a strong preference for SUMO-2/3, with little detectable sumoylation by SUMO-1. Mapping studies identified lysine 35 as the principal residue for covalent conjugation of SUMO to HPV-16 L2. We also showed that that L2 stability was maintained by sumoylation, whereas the localization of L2 was not affected.
