The results of our study showed that electroporation is feasible and effective method for delivering of plasmid DNA expressing shRNA into tumor cells in vitro, which was shown through efficient silencing of targeted reporter gene for GFP. Imaging and measurements of GFP expression levels over time in vitro showed that electroporation in combination with plasmid DNA as gene delivery vector can induce prolonged gene silencing effect in rapidly dividing LPBGFP cells.

The paper describes the development of stably transformed HepG2 cell line. Gen encoding green fluorescent protein (GFP) under the control of promoter of p21 gene was introduced into the cell by means of electroporation. Exposure of the prepared cell line to different genetoxins leads to activation of p21 promoter and consequently increased GFP expression, that can be easly measured by fluorometer. In the paper the development and validation of cell biosensor system with 4 genotoxin with known mechanisms of action was presented.
