The present disclosure provides an engineered NFAT (Nuclear factor of activated T cells)-based artificial transcription regulatory system composed of a transcription factor (TF) combined a with heterodimerization (HD) domain which enables to regulate cell activation and proliferation upon the addition of external chemical signals as regulators of heterodimerization. NFAT artificial transcription factors (TF) of the present invention can be preferenntialy used to regulate the proliferation and activation of engineered therapeutic T cells to prevent the excessive cell activation by interfering with a downstream signaling pathway. NFAT artificial transcription factors of the present disclosure can be used also in various CAR T cells.
