In this publication, we published the genome of Colorado potato beetle. As members of the i5k consortium, we have participated in gene model correction, contributed RNAseq data and manually annotated several gene families, including those used as target genes in the present study. We have used the preliminary genome sequences and the RNA-seq expression data in the selection of target genes phases of the project.

We have developed quantGenius (http://quantgenius.nib.si), an open-source web application which enables reliable quantification of data obtained with the real-time PCR method (qPCR). The quantGenius workflow interactively guides the user through data import, quality control (QC) and quantification steps. The special feature of the application is the implementation of user-guided QC-based decision support system, based on qPCR standards, that takes into account pipetting errors, assay amplification efficiencies, limits of detection and quantification of the assays as well as the control of PCR inhibition in individual samples. The tool will help scientists to obtain reliable results, which are the basis for biologically meaningful data interpretation. quantGenius together with appropriate qPCR setup was used in this project to quantify the expression of Colorado potato beetle target genes.
