Significance for science
Results of the project are in our opinion great contribution towards the discovery of molecular-genetic findings in lung, head and neck tumours. We reported novel associations between genetic changes on DNA, RNA and miRNA level and different tumour types, histological changes and clinical data. We discovered associations between miRNA and gene expression that together with target prediction serve as indirect proof of gene regulation via miRNAs. During the project we discovered alterations that differ between different types of tumours and transcription profiles specific for lung and head and neck tumour types. With the results of gene and miRNA expression analyses we elicited genes and miRNAs, which could serve as markers for specific stages, histological and morphological characteristics and associated them with clinical data of the patients. The study is contribution of molecular-genetic research of some of the most prevalent forms of lung, head and neck cancers. It is a contribution to the knowledge of gene and miRNA expression in primary tumours and contribution to the knowledge of gene and miRNAs differences in expression that are associated with the severity of the disease. With the project (regarding to the publications, presentations and citations of our work) we succeeded in characterizing novel molecular-genetic differences and stressed the need to proceed with characterization of genetic diversity in different tumour types which could lead in discovery and later use of diagnostic and treatment markers.
Significance for the country
With this project we introduced novel genomic technologies in the Department of molecular genetics, Institute of pathology. We also strengthened the collaboration with pathologists and clinicians that contributed to derivation of the project. We found and published data about the genes, which could in future serve as molecular markers of severity of the disease, of clinical differences and of histological differences and could find their use in diagnostics and treatment choices. With DNA microarrays, used in this project, was targeted analysis of specific gene-sets proved to be an appropriate method in establishing specific expression profiles. We were also successful in elucidation of genes that harbour mutations or have differences in expression and are associated with different histological or morphological tumour types. Microarrays that were constructed for the purpose of this project were successfully implemented in other, ongoing studies in the Department of Molecular genetics. With presenting the results of the project in publications and presentations we represented our department, Faculty of Medicine and Slovenian science and proved the quality, modernity and relevance of research done in the department of Molecular genetics, Faculty of Medicine, when comparing to other (bigger) research groups. miRNA regulation as the second possibility of gene expression shows on extreme importance of miRNA in tumourigenesis. With the discovery of miRNAs, that have different expression in tumours we indicated novel possible pathways of the tumourigenesis. Regulatory RNA molecules, for which we proved differences in expression according to tumour type or versus normal tissue, could lead to new studies expecially functional studies of the miRNAs which will result in discovery of miRNA that could serve as inhibitors or activators of the genes, that are because of their deregulation involved in formation or progression of the disease.
