Significance for science
The obtained results are relevant to a number of important questions in mammalian and vertebrate regulatory genomics. We have elucidated the origin of transcriptional repressors (why, when, where and how), their evolution, functional diversifications and expansions that led to the huge diversification of regulatory network in land vertebrates (tetrapods). The phylogenomic analysis of transcriptional repressor genes in the oldest mammalian genomes, such as monotremes, marsupials and basal placental superorders (Afrotheria and Xenarthra)), provided the definitive answer on the origin and evolution of transcriptional repressors in mammals. The acquisition of lineage- and species-specific transcriptional repressors may hold the key to understanding gene expression and biological differences that make mammals distinct from their closest relatives. The mechanisms by which transcriptional regulatory networks of transcriptional repressors in tetrapods originate, evolve and expand have been elucidated. The particular characteristics of DNA binding (ZNF) and effector (KRAB and SCAN) domains that were responsible for the origin of transcriptional repressors in tetrapods and for their massive increase in number and complexity have been elucidated. The role and impact of de novo exonization of the SCAN dimerization domain in the basal tetrapods have been elucidated. The mechanisms involved in the origin and expansion of the huge family of transcriptional repressors have been elucidated. The obtained results are important for various scientific fields: comparative genomics, phylogenomics, mammalian and vertebrate biology, and regulatory and functional evolution.
Significance for the country
Our original research in the field of genome research has enabled the introduction of new methods and technologies in the fields of genome biology, »in silico« biology, bioinformatics and molecular evolution in Slovenia. We acquire new and practical knowledge, information and skills in the field of comparative and evolutionary genomics. The consequence of our current research work is that Slovenia is not excluded from the field of genome research. The results of our original research are published in leading scientific journals and presented at international scientific meetings. Theoretical and practical knowledge from the fields of genomics, proteomics and molecular evolution is transferred to the students and young researchers at the University of Ljubljana and IJS. Diploma students and young researchers are trained and educated in the field of genomics; this newly obtained knowledge is transferred into the pharmaceutical industry and to other research institutions. Research project was harmonized with the national research and development programme and with the thematic priorities within the public call and belongs to the first thematic priority: Life sciences, genomics and biotechnology for health. This project was connected to the research on the comparative genomics of vertebrates, which is the part of research programme P1-0207.
