Abstract
Purification and concentration of plant viruses by chromatography would shorten the purification and/or concentration of different viruses. This area is expected to develop very fast in next few years, especially because of the potential usage of recombinant viruses as vectors in gene therapy. Chromatographic supports applicable for fast and efficient purification will become indispensable. Our preliminary tests showed that monolithic CIM® supports have a lot of advantages in comparison to other classical chromatographic supports, above all they enable fast and efficient concentration and separation.This project will serve us as a study of virus behavior on chromatographic supports. Because of the safety reasons our model will be plant viruses, which represent no hazard to environment or personnel. First we will propagate model plant virus in a suitable propagation plant, remove the plant material clarification and continue purification and concentration on monolithic chromatographic support (CIM®). Chromatographic conditions will be optimized as well as the structure of the pores of the chromatographic support. We will use ion exchangers, and where possible, also affinity monolithic CIM® columns.The results of this project will serve as a basis of preparing different products for purification and concentration of different plant viruses. The applicability of these products will be important especially for improvement of detection methods (e.g. ELISA, PCR) of viruses in different samples, above all in samples with low concentration of viruses (e.g. potable water). The knowledge which we will gain will be used for development of new monolithic CIM® supports and the methods for purification of virus vectors. Also we will use it for the development of supports and methods for improvement of detection methods of human viruses, especially in potable water.
